How does the medication work?
The medication belongs to a group of medicines called antidepressants. The medication has two
enantiomeric racemic forms. Anti-depressive effect of the medication is related to enhancement of
neurotransmitters activity. Vanlafaxin and its active metabolite O-dismethylvenlafaxine are inhibitors of serotonin and noradrenalin reuptake. Venlafaxine also inhibits the reuptake of dopamine. By increasing the amount of these neurotransmitters in the synaptic gap, Venlafaxine helps to relieve the symptoms of depressive disorders. Venlafaxine decreases beta-adrenergic reactions and does not possesses an affinity to m- and n-choline receptors, histamine H1-receptors and a1- adrenergic receptors of the brain. Venlafaxine does not inhibits MAO activity.
Why is Effexor prescribed?
Effexor is used in the treatment of depressive disorders (accompanied by anxiety), treatment and prophylaxis of relapses
When Effexor should not be taken?
You should not take Effexor if any of the following conditions apply to you:
- Severe kidney dysfunction (creatinine clearance less than 10ml/min)
- Severe liver dysfunction
- Concomitant use of MAO applications
- Children and adolescents under 18 years of age
- Pregnancy
- Breastfeeding
The medication should be used with caution in recent heart attacks, unstable angina, heart failure, diseases of the coronary arteries, ECG changes(including prolonged QT), disorders of the electrolyte balance, hypertension, increased heart rate, seizures in anamnesis, intra ocular pressure, closed angle glaucoma, maniac conditions in anamnesis, tendency to bleedings, decreased body's weight.
What side effects are possible with the medicine ?
- Systemic side effects: weakness, fatigability, fever
- Digestive system: decreased appetite, constipation, nausea, vomiting, dry mouth, stomach pain,
- digestive bleedings, hepatitis
- CNS: hyper-irritability, insomnia, agitation, paresthesia, tremor, ataxia,
- Cardiovascular system: hypertension, dilation of the blood vessels (hot flashes), increased heart rate, faint, arrhythmia, prolonged QT interval, ventricular tachycardia, ventricular fibrillation
- Sense organs: accommodation disorders, mydriasis, vision disorders, tinnitus, taste changes
- Haemopoietic system: hemorrhage into the skin and mucous linings, thrombocytopenia
- Dermatological reactions: increased sweating, photosensibilisation, alopecia
- Metabolism: high cholesterol blood levels, decreased body weight, low potassium blood levels, increases blood levels of prolactin.
- Urogenital system: urine retention, ejaculation problems, erection problems, anorgasmia, decreased libido, disorders of the menstrual cycle, menorrhagia, galactorrhea
- Apparatus system: arthralgia, myalgia, muscular spasms.
- Allergic reactions: skin rash, itching, angio edema, hives, erythema.
- Sudden cessation of therapy may cause withdrawal symptoms

What are possible drug to drug interactions associated with Effexor?
Effexor should not be taken in combination with:
- Blood thinners (anticoagulants and antiplatelets) including aspirin and warfarin
- NSAID's
- HIV treatment (indinavir)
- Antimycotics (ketoconazole)
- Antibiotics (linezolid)
- Lithium preparations
- MAO inhibitors
- Other antidepressants
- Narcotic analgesics (tramadol)
- Triptans
- Tryptophan
- St.John's wort
- Antipsychotics (Haloperidol)
Let your doctor know if you are taking any of the above mentioned medicines or other preparations including herbals, vitamins and food supplements as they may change the effectiveness or increase the risk of Effexor side effects.
