How does the medication work?
Duloxetine belongs to a group of medicines called serotonin and norepinephrin reuptake inhibitors. By inhibiting the reuptake of these neurotransmitters duloxetine increases their amount in the synaptic gap and thus improve the intraneuronal transfer of impulses. All of these helps to decrease or eliminate the symptoms of depression. The medication does not possess an affinity to histamine, dopamine, cholinergic and adrenergic receptors. Duloxetine possesses a central mechanism of suppression the pain syndrome, this leads to increase of threshold of pain sensitivity in pain syndrome of neuropathic etiology.
Why is Duloxetine prescribed?
Duloxetine is used in the treatment of the following conditions:
- Depression
- Pain associated with diabetic neuropathy
When Duloxetine should not be taken?
Duloxetine is contraindicated in the following conditions:
- Hypersensitivity to duloxetine or other components of the medication
- Concomitant application with MAO inhibitors
- Closed-angle glaucoma
- Pregnancy
- Breastfeeding
- Severe liver diseases
- Severe kidney diseases
Duloxetine should be used with caution in aggravation of manic/hypomaniac conditions, epilepsy, mydriasis, liver or kidney dysfunction, in patients with tendency to suicidal thoughts or attempts.
What side effects are possible with the medicine ?
Duloxetine side effects include:
- CNS: dizziness, vision problems, headache, tremor, blurred vision, anxiety, yawning, in rare cases glaucoma, mydriasis, agitation, disorientation.
- Digestive system: dry mouth, nausea, constipation, diarrhea, vomiting, decreased appetite, taste changes, liver dysfunction, in very rare cases: hepatitis, jaundice, increased activity of liver's enzymes, belching.
- Apparatus system: muscular tensity, bruxism.
- Cardiovascular system: increased heart rate, orthostatic hypotension, increased heart rate, paresthesia
- Urogenital system: anorgasmia, decreased libido, retention and ejaculation disorders, erectile dysfunction, difficulties with urination. In very rare case- nycturia.
If any of these side effects occur during the treatment seek medical attention immediately.
