How does the medication work?
Antidiabetic hypoglycemic drug for oral administration, derivative of thiazolidinediones. Selectively stimulates the nuclear gamma receptors activated by peroxisome proliferator-activated receptors-gamma (PPAR-gamma agonist). Activation of the nuclear PPAR-gamma modulates the transcription of genes that are sensitive to insulin and are involved in control of glucose level, lipid metabolism in the liver, in the adipose and muscle tissues.
Actos lowers insulin resistance of peripheral tissues and liver and, as a result, increases the insulin-dependent glucose consumption and reduces glucose release from the liver. In type II diabetes reduced insulin resistance under the action Actos causes a decrease in the concentration of glucose, insulin level and glycated hemoglobin in blood plasma. In monotherapy and in combination with sulfonylureas, metformin, or insulin Actos improves control of blood sugar.
In type II diabetes with violation of lipid metabolism, Actos reduces the triglycerides level and raises HDL without altering LDL and total cholesterol in the blood plasma.
Why is Actos prescribed?
- diabetes mellitus (II type) - as monotherapy or in combination with derivatives of sulfonylurea, metformin, or insulin if diet therapy, exercises, and monotherapy with one of the above drugs were inefficient
When Actos should not be taken?
- Hypersensitivity to pioglitazone or to one of the components of Actos
- Insulin-dependent diabetes mellitus (type I)
- Diabetic ketoacidosis
- Acute liver disease
- If the level of alanine aminotransferase is higher than normal in 2.5 times and more
- Pregnancy and breastfeeding
- With caution: edema, moderate increase of liver enzymes level, pediatric and adolescent patients
What side effects are possible with Actos ?
- Hypoglycemia
- Edema
- Anemia, decreased hemoglobin and hematocrit
- Increased alanine aminotransferase / ALT and creatine phosphokinase / CPK in blood plasma.
If any of these side effects occur during the treatment seek medical attention immediately.

Actos is known to interact with the following groups of drugs:
- derivatives of sulfonylurea, metformin and insulin

- antifungal agents
